目的 :探讨胰岛素样生长因子 -1受体 (IGF 1R)mRNA在低氧大鼠肺小动脉壁的变化。方法 :采用原位杂交方法检测低氧性肺动脉高压大鼠和正常大鼠肺小动脉壁IGF 1RmRNA的表达 ,用图像分析技术检测肺小动脉的形态改变和半定量技术检测IGF 1RmRNA的表达强度。结果 :低氧性肺动脉高压大鼠肺小动脉管壁增厚、管腔狭窄 ,反映管壁增厚的两个指标MT %和MA %均显著高于正常对照组 (P <0 .0 5 )。正常对照组大鼠肺小动脉壁IGF 1RmRNA有微弱表达 ,低氧组肺小动脉壁IGF 1RmRNA表达呈强阳性 ,低氧组表达强度显著大于正常对照组 (P <0 .0 0 1)。结论 :低氧情况下 ,肺小动脉壁IGF 1RmRNA表达水平与低氧性肺血管重建密切相关
AIM: To investigate the changes of mRNA of insulin-like growth factor-1 receptor (IGF-1R) in the pulmonary arterioles of hypoxic rats. Methods: The expression of IGF1R mRNA in pulmonary arterioles of hypoxic pulmonary hypertensive rats and normal rats was detected by in situ hybridization. The morphological changes of pulmonary arterioles were detected by image analysis and the expression of IGF1R mRNA was detected by semi-quantitative analysis. Results: The pulmonary arteriole thickened and stenosed lumen in hypoxic pulmonary hypertensive rats showed that MT% and MA% were significantly higher than those in normal control group (P <0.05) . The expression of IGF-1R mRNA in the pulmonary arterioles wall was weakly positive in the normal control group. The expression of IGF-1R mRNA in the hypoxic group was stronger than that in the normal control group (P <0.01). CONCLUSIONS: Under hypoxic conditions, the expression of IGF-1R mRNA in pulmonary arterioles is closely related to hypoxic pulmonary vascular remodeling
