目的研究COX_2c、_erbB_2、Ki_67在星形胶质细胞增生及星形胶质细胞瘤中的表达,并探讨三者在鉴别反应性与肿瘤性星形胶质细胞方面的作用。方法利用组织芯片技术及PV6000通用型二步法免疫组化方法检测COX_2c、_erbB_2、Ki_67在正常脑组织、星形胶质细胞增生、低/高级别星形胶质细胞瘤中的表达。结果正常组织中COX_2c、_erbB_2、Ki_67表达均为(-);增生组中阳性表达率分别为28.9%、37.8%和22.2%,与正常组比较,COX_2、Ki_67表达差异不显著,c_erbB_2差异显著(P<0.05);低级别肿瘤组中三者阳性表达率分别为68.1%、63.8%和70.2%,与增生组比较,均差异显著(P<0.05);高级别肿瘤组中三者阳性表达率分别为88.1%8、3.3%和95.2%,与低级别肿瘤组比较,均差异显著(P<0.05)。COX_2c、_erbB_2与组织学分级密切相关(r值分别为0.989和0.988)。结论COX_2c、_erbB_2、Ki_67有可能成为鉴别星形胶质细胞增生与低级别星形胶质细胞瘤的客观指标,且COX_2、c_erbB_2在星形胶质细胞瘤的发生、发展过程中有重要作用。
Objective To study the expression of COX_2c, _erbB_2 and Ki_67 in astrocytoma and astrocytoma, and to explore the role of the three in differentiating between reactive and neoplastic astrocytes. Methods The expression of COX_2c, _erbB_2 and Ki_67 in normal brain tissue, astrocytosis and low / high grade astrocytomas were detected by tissue microarray and PV6000 universal two-step immunohistochemistry. Results The positive expression rates of COX_2c, _erbB_2 and Ki_67 in normal tissues were (-). The positive rates of COX_2, Ki_67 were 28.9%, 37.8% and 22.2% respectively in normal group P <0.05). The positive rates of the three in the low-grade tumor group were 68.1%, 63.8% and 70.2%, respectively, which were significantly different from those in the proliferative group (P <0.05) Respectively 88.1% 8,3.3% and 95.2%, compared with the low-grade tumor group, the difference was significant (P <0.05). COX_2c and _erbB_2 were closely related to histological grade (r = 0.989 and 0.988, respectively). Conclusions COX_2c, _erbB_2 and Ki_67 may be objective indicators for differentiating astrocytoma from low grade astrocytoma. COX_2 and c_erbB_2 may play an important role in the development of astrocytoma.
