目的:筛选板蓝根中抗流感病毒的活性成分。方法:采用超滤质谱技术,筛选板蓝根中具有抗流感病毒的活性成分,并以体外神经氨酸酶活性实验进行验证。结果:通过超滤质谱筛选并鉴定出与神经氨酸酶结合的成分主要是精氨酸、告依春和腺苷,其结合常数分别为(36.23±1.12)%,(32.54±1.02)%,(9.38±0.47)%;该结果与体外酶活性实验结果一致。精氨酸、告依春的IC50分别为(1.16±0.02),(1.20±0.02)g·L-1,腺苷的IC50大于500 g·L-1。结论:精氨酸和告依春可能是板蓝根抗流感病毒的活性成分,为板蓝根质量评价指标的选择提供了科学依据;亦为中药药效物质的发现提供了一种适宜技术。
OBJECTIVE: To screen anti-influenza virus active ingredients in Radix Isatidis. Methods: The anti-influenza virus active ingredients in Radix Isatidis were screened by ultrafiltration mass spectrometry and verified by in vitro neuraminidase activity test. Results: The results showed that the binding constants of neuraminidase and arginine were (36.23 ± 1.12)%, (32.54 ± 1.02)%, (9.38 ± 0.47)%, respectively. The result was consistent with the in vitro enzymatic activity. The IC50 of arginine and eosin were (1.16 ± 0.02) and (1.20 ± 0.02) g · L-1, respectively, and the IC50 of adenosine was more than 500 g · L-1. Conclusion: Arginine and Yichunchun may be the active ingredients of the anti-influenza virus of Radix Isatidis, which provides a scientific basis for the selection of the quality evaluation index of Radix Isatidis. It also provides a suitable technique for the discovery of the effective substances of Radix et Rhizoma.
