目的 研究APP17肽对D 半乳糖脑病小鼠海马神经元凋亡的影响。 方法 用D 半乳糖制备脑老化模型 ,皮下注射APP17肽治疗D 半乳糖脑老化小鼠 ,3个月后取脑组织做Bcl 2、Bax、CREB、AKt、AIF免疫组织化学染色。 结果 D 半乳糖脑病小鼠海马内Bax、AIF阳性反应神经元胞浆与突起深染 ,而正常小鼠及APP17肽保护的D 半乳糖小鼠海马阳性细胞数目少 ,染色淡 ;D 半乳糖小鼠海马内Bcl 2、CREB、AKt阳性细胞数目少 ,染色淡 ;而正常小鼠及APP17肽保护的D 半乳糖小鼠海马阳性反应神经元胞浆与突起深染。 结论 促进凋亡的Bax、AIF在D 半乳糖小鼠海马表达增加 ;抑制凋亡的Bcl 2、CREB、AKt在D 半乳糖小鼠海马表达降低。APP17肽能影响D 半乳糖脑病小鼠脑内Bcl 2、Bax、CREB、AKt、AIF的表达 ,使之接近正常
Objective To study the effect of APP17 peptide on apoptosis of hippocampal neurons in D-galactosemicroglia mice. Methods Cerebral aging model was induced by D - galactose and APP17 peptide was injected subcutaneously into mice with aging of D - galactose brain. Three months later, the brain tissues were taken for Bcl - 2, Bax, CREB, AKt and AIF immunohistochemical staining. Results The numbers of Bax and AIF positive neurons in D-galactocerebellar mice hippocampus were deep-stained with protuberances, while the numbers of hippocampal positive cells in D-galactose mice protected by normal mice and APP17 peptide were small, In the hippocampus of rats, the number of Bcl-2, CREB and AKt positive cells was less and the staining was faint. However, the hippocampal positive neurons of D-galactose mice protected by normal mice and APP17 peptide had deep staining. Conclusion Apoptosis of Bax and AIF increased in the hippocampus of D-galactose-induced mice. The expression of Bcl-2, CREB and AKt decreased in the hippocampus of D-galactose induced mice. APP17 peptide can affect the expression of Bcl 2, Bax, CREB, AKt and AIF in the brain of D-galactosemicrotrophic mice and make it close to normal
