目的探讨甲醛对小鼠源性巨噬细胞(RAW264.7细胞)DNA的损伤作用。方法用0,20,40,80,160,320μmol/L甲醛处理RAW264.7细胞1 h。采用单细胞凝胶电泳技术检测甲醛对RAW264.7细胞DNA的损伤作用。结果甲醛剂量为20,40,80,160μmol/L时,RAW264.7细胞DNA的迁移率和尾长分别为33.0%,49.7%,82.3%,38.3%和11.2,22.6,30.1,18.9μm,与空白对照组4.3%和9.7μm比较,差异有统计学意义(P<0.05);当甲醛浓度达320μmol/L时,细胞DNA的迁移率和尾长分别为6.3%和9.3μm,与对照组比较,差异无统计学意义(P>0.05)。结论低浓度甲醛可造成RAW264.7细胞DNA的损伤。
Objective To investigate the effect of formaldehyde on the DNA damage of mouse-derived macrophages (RAW264.7 cells). Methods RAW264.7 cells were treated with 0, 20, 40, 80, 160 and 320 μmol / L formaldehyde for 1 h. Single cell gel electrophoresis was used to detect DNA damage of RAW264.7 cells. Results The DNA migration rate and tail length of RAW264.7 cells were 33.0%, 49.7%, 82.3%, 38.3% and 11.2, 22.6, 30.1 and 18.9 μm, respectively, at the doses of 20,40,80 and 160μmol / (P <0.05). When the formaldehyde concentration reached 320μmol / L, the cell DNA migration rate and tail length were 6.3% and 9.3μm, respectively. Compared with the control group, The difference was not statistically significant (P> 0.05). Conclusion Low concentration of formaldehyde can cause DNA damage in RAW264.7 cells.
