目的:观察α干扰素对鼻咽癌细胞株CNE2的体外生长抑制和诱导凋亡作用,为其在抗肿瘤治疗方面的应用提供理论依据。方法:α干扰素治疗组和未治疗组分别采用免疫印记检测磷酸化AKT和caspase-3蛋白水平、XTT检测癌细胞生长和流式细胞术分析癌细胞凋亡。结果:α干扰素与CNE2细胞株共同培养48h后,磷酸化AKT蛋白水平明显低于CNE2细胞组,而caspase-3蛋白水平明显高于CNE2组;XTT试验显示α干扰素明显抑制CNE2细胞系的的生长,这种抑制作用呈现时间和浓度依赖性;AnnexinV/PI双染流式细胞检测α干扰素与CNE2细胞系共育48h后细胞的早期凋亡率23.42%,而对照组的早期凋亡率仅为4.5%(P<0.05)。结论:α干扰素诱导鼻咽癌细胞凋亡,从而抑制其体外生长增殖,具有抗肿瘤作用。
OBJECTIVE: To observe the inhibitory effect of interferon-alpha on nasopharyngeal carcinoma cell line CNE2 in vitro and to induce apoptosis, and to provide a theoretical basis for its application in anti-tumor therapy. Methods: The levels of phosphorylated AKT and caspase-3 were detected by Western blotting in the treatment group and the untreated group respectively. The growth of cancer cells was detected by XTT and the apoptosis of cancer cells was analyzed by flow cytometry. Results: After IFNα and CNE2 cells co-cultured for 48h, the phosphorylated AKT protein level was significantly lower than that of CNE2 cells and caspase-3 protein level was significantly higher than that of CNE2 cells. XTT assay showed that interferon α significantly inhibited CNE2 cell line . The early apoptotic rate was 23.42% after AnnexinV / PI double staining flow cytometry assay for interferon alpha and CNE2 cells for 48h, while the early apoptosis in control group The rate was only 4.5% (P <0.05). CONCLUSION: Alpha interferon induces the apoptosis of nasopharyngeal carcinoma cells and inhibits the proliferation and proliferation of nasopharyngeal carcinoma cells in vitro with anti-tumor effect.
