目的探讨大黄素对人胃癌BGC-823细胞增殖和迁移的影响。方法采用四甲基偶氮唑盐(MTT)比色法测定不同浓度大黄素对体外培养人胃癌BGC-823细胞增殖的影响;采用流式细胞术测定大黄素对人胃癌BGC-823细胞的细胞周期影响;采用划痕损伤实验测定大黄素对人胃癌BGC-823细胞迁移的影响。结果 MTT法显示,大黄素能抑制人胃癌BGC-823细胞増殖,呈现浓度依赖性;流式细胞术显示,人胃癌BGC-823细胞的细胞周期主要停滞在G0/G1期;划痕损伤实验表明,人胃癌BGC-823细胞迁移受到明显的抑制。结论大黄素对人胃癌BGC-823细胞的增殖和迁移具有明显的抑制作用,且呈浓度依赖性,并可使人胃癌BGC-823细胞增殖周期停滞于G0/G1期。
Objective To investigate the effects of emodin on the proliferation and migration of human gastric cancer cell line BGC-823. Methods MTT assay was used to determine the effect of different concentrations of emodin on the proliferation of human gastric cancer cell line BGC-823. Flow cytometry was used to determine the effect of emodin on the proliferation of human gastric cancer BGC-823 cells Cycle impact; Scratch damage test was used to determine the emodin on human gastric cancer BGC-823 cell migration. Results MTT assay showed that emodin could inhibit the proliferation of human gastric cancer cell line BGC-823 in a concentration-dependent manner. Flow cytometry showed that the cell cycle of human gastric cancer cell line BGC-823 mainly arrested in G0 / G1 phase. , Human gastric cancer BGC-823 cell migration was significantly inhibited. Conclusion Emodin can significantly inhibit the proliferation and migration of human gastric cancer cell line BGC-823 in a concentration-dependent manner and arrest the proliferation of human gastric cancer cell line BGC-823 in G0 / G1 phase.
