目的:研究四方藤60%乙醇提取物对类风湿关节炎模型大鼠关节滑膜组织的作用及机制。方法:采用CⅡ胶原加氟氏完全佐剂法诱导类风湿关节炎大鼠模型,测定大鼠滑膜组织中TNF-α、NF-кB的表达,免疫组化法检测滑膜组织中内皮细胞黏附因子-1(ICAM-1)的表达,并对滑膜进行病理学检测。结果:与模型组比较,四方藤提取物能显著降低模型大鼠滑膜组织中炎症因子TNF-α、NF-кB的表达(P<0.01)及关节滑膜组织中ICAM-1的表达(P<0.01);病理检测表明,四方藤提取物能显著改善类风湿关节炎大鼠的关节滑膜病变。结论:四方藤提取物可对类风湿关节炎大鼠具有显著的治疗作用,其机制可能与下调滑膜组织中TNF-α、NF-кB、ICAM-1的表达有关。
Objective: To study the effect and mechanism of 60% ethanol extract of Tetrahedron vine on synovial tissue of rheumatoid arthritis rats. Methods: The model of rheumatoid arthritis was induced by CⅡcollagen and complete Freund’s adjuvant. The expression of TNF-α and NF-κB in synovial tissue of rats were measured. The adhesion of endothelial cells in synovial tissue was detected by immunohistochemistry 1 (ICAM-1) expression, and synovial pathological examination. Results: Compared with the model group, Sifang vine extract could significantly decrease the expression of TNF-α and NF-κB (P <0.01) and the expression of ICAM-1 in synovial tissue (P <0.01). Pathological examination showed that Sifang vine extract can significantly improve the synovial lesions of rheumatoid arthritis rats. Conclusion: Sifang rattan extract may have a significant therapeutic effect on rheumatoid arthritis rats. The mechanism may be related to down-regulating the expression of TNF-α, NF-κB and ICAM-1 in synovial tissue.
