目的采用HPLC法同时测定宜宾产蒲公英中咖啡酸和绿原酸的含量。方法色谱柱为Waters symmetry shieldTM C18柱(250mm×4.6mm,5μm),流动相为乙腈-0.1%磷酸(10∶90),检测波长328nm,流速1.0mL·min-1。结果咖啡酸、绿原酸的线性范围分别为0.08~0.80μg(r=0.9992)、0.04~0.40μg(r=0.9994);平均回收率分别为101.4%(RSD=2.9%)、100.0%(RSD=3.2%)。结论绿原酸的含量为0.011%~0.052%,咖啡酸的含量为0.035%~0.069%,咖啡酸的含量远高于2005年版《中国药典》规定的0.02%的限度。所用方法准确、可靠,可用于蒲公英的质量控制。
Objective To determine the content of caffeic acid and chlorogenic acid in dandelion from Yibin by HPLC. Methods The column was a Waters symmetry shieldTM C18 column (250mm×4.6mm, 5μm). The mobile phase consisted of acetonitrile-0.1% phosphoric acid (10∶90). The detection wavelength was 328nm and the flow rate was 1.0mL·min-1. Results The linear ranges of caffeic acid and chlorogenic acid were 0.08-0.80 μg (r=0.9992) and 0.04-0.40 μg (r=0.9994), respectively; the average recoveries were 101.4% (RSD=2.9%) and 100.0% (RSD, respectively). =3.2%). Conclusion The content of chlorogenic acid is 0.011%-0.052%, the content of caffeic acid is 0.035%-0.069%, and the content of caffeic acid is much higher than the limit of 0.02% prescribed in the 2005 edition of the Chinese Pharmacopoeia. The method used is accurate and reliable and can be used for quality control of dandelion.
