目的:建立新疆阿魏抗溃疡提取物中阿魏酸的HPLC含量测定方法。方法:运用高效液相色谱法(HPLC法)对新疆阿魏抗溃疡提取物中阿魏酸的含量进行测定并根据10批样品的测试结果确定提取物中阿魏酸的含量限度。采用KromasilODS-1 C_(18)柱(250 mm×4.6 mm,0.5μm),流动相为乙腈-0.085%磷酸溶液(17:83),流速为1 ml/min,进样量为10μl,柱温为35℃,检测波长为322 nm。结果:线性范围为4.04~161.60μg/ml,相关系数为0.999 9,回收率99.47%,RSD=1.33%(n=9)。结论:HPLC法快捷、准确、操作简便、灵敏、重现性好,适用于阿魏酸的含量测定,为阿魏抗溃疡提取物开发利用提供了实验基础。
Objective: To establish an HPLC method for the determination of ferulic acid in extracts of anti-ulcers from Xinjiang ferulae. Methods: The contents of ferulic acid in extracts of the anti-ulcer extracts from Xinjiang Ferula were determined by high performance liquid chromatography (HPLC) and the limits of ferulic acid content in extracts were determined according to the results of 10 batches of samples. A Kromasil ODS-1 C18 column (250 mm×4.6 mm, 0.5 μm) was used. The mobile phase was acetonitrile-0.085% phosphoric acid solution (17:83). The flow rate was 1 ml/min, and the injection volume was 10 μl. It is 35°C and the detection wavelength is 322 nm. Results: The linear range was from 4.04 to 161.60 μg/ml, the correlation coefficient was 0.999 9, the recovery rate was 99.47%, and RSD was 1.33% (n=9). Conclusion: HPLC method is rapid, accurate, easy to operate, sensitive and reproducible. It is suitable for the determination of ferulic acid content, and provides an experimental basis for the development and utilization of anti-ulcer extracts of A. ferrum.
