目的了解中山地区蜚蠊中沃尔巴克氏体的感染状况及其基因型别。方法于2015年6月至2016年7月,采集广东省中山市的常见蜚蠊种类,通过扩增沃尔巴克氏体的wsp基因并测序判断其是否感染,同时进行系统进化分析;用配对设计t检验分析不同种类雌雄个体感染率差异是否有统计学意义。结果共检测12种154只蜚蠊样本,其中5种42只蜚蠊检出感染沃尔巴克氏体,个体感染率为27.27%,且雌雄感染率差异无统计学意义(|t|=0.542,|t|0.01)。共获得8个wsp基因单倍型序列,其分别属于wAlbB、wAlbD和wAlbF型沃尔巴克氏体。结论首次发现维丽斯璐蠊、中华玛啦蠊、姬蠊科sp.、玛蠊属sp.1和玛蠊属sp.2携带沃尔巴克氏体,其基因型属于wAlbB、wAlbD和wAlbF型,且可能在维丽斯璐蠊体内检测到1种新的wAlbF基因型的沃尔巴克氏体。
Objective To understand the infection status and genotypes of Wolbachia in cockroaches in Zhongshan area. Methods From June 2015 to July 2016, common cockroaches were collected from Zhongshan City, Guangdong Province. Wsp genes of Wolbachia were amplified and sequenced to determine whether they were infected. Phylogenetic analysis was also performed. t test analysis of different types of male and female individual differences in infection rates were statistically significant. Results A total of 12 samples of 154 cockroaches were detected. Among them, 42 cockroaches infected by 5 cockroaches were found to be infected with Wolbachia. The individual infection rate was 27.27%, and there was no significant difference between male and female infection rates (| t | = 0.542, | t | 0.01). A total of 8 wsp gene haplotypes were obtained, which belong to wAlbB, wAlbD and wAlbF type Wolbachia. CONCLUSIONS It was first found that Wolbachia, wapiti, wAlbD and wAlbF genotypes belonged to Wolbachia, Chrysopa sinica, Blattaceae sp., Blattaceae sp.1 and Blattaceae sp.2 , And one new Wolbachia gene of wAlbF genotype may be detected in Veronese cockroach.
