目的:研究牛蒡ITS序列与牛蒡子药材质量相关性。方法:采集全国26个不同产地的牛蒡子药材,采用PCR扩增后测定ITS序列,HPLC法对牛蒡子药材中牛蒡苷含量进行测定,应用ClustalX(1.81),Mage 4.0,SPSS13.0等统计软件进行遗传多样性、基因分型、相关性等分析。结果:测得了26份牛蒡的ITS序列全长,在GenBank中注册,获得登记号,测定牛蒡子药材中牛蒡苷含量、千粒重,统计分析牛蒡的基因分型与药材质量具有相关性。结论:为揭示牛蒡子道地药材的分子形成机制奠定了基础。
Objective: To study the correlation between burdock ITS sequence and Arctii quality. Methods: Arctiaceae was collected from 26 burdock plants in different areas of China. ITS sequence was determined by PCR amplification. The content of arctiin in Arctium lappa L. was determined by HPLC. Statistical software such as ClustalX (1.81), Mage 4.0, SPSS 13.0 Genetic diversity, genotyping, correlation and other analysis. Results: The total length of ITS sequences of 26 burdocks was measured and registered in GenBank. The curcumin content and thousand-grain weight were determined in Arctium lappa L., and the genotypes of Burdock were statistically analyzed. Conclusion: It laid the foundation for revealing the molecular mechanism of Arctii officinalis.
