目的以秀丽隐杆线虫为模式生物制备人源脑红蛋白(human neuroglobin,hNgb)转基因线虫,初步探讨外源性hNgb对线虫寿命和产卵率的影响。方法通过显微注射方法制备hNgb转基因线虫,使用蛋白印迹和免疫组织化学进行鉴定。使用野生型N2和增强型绿色荧光蛋白(EGFP)转基因线虫作为对照组,测量转基因线虫的寿命和产卵率。结果成功获得hNgb转基因株系线虫,其寿命和产卵率与野生型N2和EGFP转基因线虫相比,差异无统计学意义。结论外源性hNgb转基因不影响线虫的发育和正常生理功能,可用于Ngb功能的深入研究。
OBJECTIVE: To prepare human neuroglobin (hNgb) transgenic nematode by using Caenorhabditis elegans as a model organism to investigate the effects of exogenous hNgb on nematode life expectancy and oviposition rate. Methods The hNgb transgenic nematode was prepared by microinjection method and identified by Western blotting and immunohistochemistry. Wild-type N2 and enhanced green fluorescent protein (EGFP) transgenic nematodes were used as controls to measure the longevity and oviposition rate of transgenic nematodes. Results The hNgb transgenic nematode was successfully obtained. The difference of the longevity and spawning rate between the two groups was not statistically significant compared with wild-type N2 and EGFP transgenic nematodes. Conclusion Exogenous hNgb transgene does not affect the development and normal physiological function of nematode, which can be used for the further study of Ngb function.
