目的:观察氟哌利多(Dro,多巴胺拮抗剂)与电针(EA)合用后前阿黑皮原(POMC)和前强啡肽原(PPD)mRNA的表达.方法:大鼠经处理后取脑与脊髓切片,采用原位杂交技术观察.结果:EA后10h,POMCmRNA表达增强,Dro+EA后,POMCmRNA表达进一步增强.PPDmRNA的表达在脊髓被EA增强,Dro+EA进一步促进其表达;在脑内,EA或Dro+EA后PPDmRNA的表达在尾壳核,伏核,下丘脑等脑区无明显变化,在视上核则减弱.结论:氟哌利多与电针合用促进了中枢POMCmRNA和脊髓PPDmRNA的表达,对脑内PPDmRNA的表达则有减弱作用(视上核)或无明显影响.
OBJECTIVE: To observe the expression of pro-opioid (POMC) and prepro-proendorphin (PPD) mRNAs after droperidol (Dro, dopamine antagonist) and electroacupuncture (EA) Methods: After treatment, the brain and spinal cord sections were taken and observed by in situ hybridization. Results: The expression of POMC mRNA tended to be enhanced 10 h after EA and the expression of POMC mRNA was further enhanced after Dro + EA. The expression of PPD mRNA in the spinal cord was enhanced by EA, and Dro + EA further promoted the expression of PPDmRNA. The expression of PPDmRNA in the brain, EA or Dro + EA had no significant changes in the caudate putamen, nucleus accumbens, hypothalamus and other brain regions, but decreased in the supraoptic nucleus. Conclusion: Combination of droperidol and electroacupuncture can promote the expression of PPDmRNA in central POMC and spinal cord, but have no effect on the expression of PPDmRNA in the brain (supraoptic nucleus).
