本文利用免疫沉淀、双相凝胶电泳技术分析了某些DR位点纯合子细胞DR抗原电泳图谱。待检细胞用~3H-亮氨酸生物合成性标记,提取膜蛋白,对抗DR单态型抗血清做免疫沉淀,沉淀物用双相凝胶电泳进行分析。结果表明,DR抗原α肽链电泳图单一,各DR型间无差异:β肽链电泳图谱因DR型别而异,是区分DR型别的主要依据。此外,2例DR2纯合子细胞图谱明显不同,提示为两个不同的亚型。
In this paper, immunoprecipitation, two-phase gel electrophoresis analysis of some DR site homozygous DR antigen electrophoresis. The cells to be tested were labeled with ~ 3H-leucine biosynthesis, membrane proteins were extracted, immunoprecipitated against DR monotype antiserum, and the pellet was analyzed by two-phase gel electrophoresis. The results showed that there was no difference between DR antigen α peptide electrophoresis patterns and DR types. The β-peptide electrophoresis pattern was different from DR type, which was the main basis for distinguishing DR type. In addition, two DR2 homozygous cell lines were significantly different, suggesting two different subtypes.
