采用盐酸水解、邻苯二甲醛- 氯甲酸芴甲酯(OPA- FMOC) 柱前衍生反相高效液相色谱法测定了经碱性蛋白酶脱蛋白处理后的天然胶乳中可溶性蛋白质的氨基酸含量。从16 种氨基酸的变化考察了所选酶对乳胶中可溶性蛋白质的水解性能, 以氨基酸总量变化评价脱蛋白的效果。结果表明, 所选用的碱性蛋白酶对胶乳中的可溶性蛋白的肽键都能水解, 在水解过程中蛋白质的氨基酸的组成是变化的。 该法对胶乳中可溶性蛋白质的检出限达1 .5 mg/ L( 进样5 μL, S/ N= 2) , 结果不受蛋白质中氨基酸组成变化的影响。 对牛血清白蛋白的氨基酸组成分析结果表明, 该法准确度高。
Amino acid content of soluble protein in natural rubber latex deproteinized by alkaline protease was determined by pre-column derivatization with OPA-FMOC using hydrochloric acid hydrolysis. The hydrolytic properties of the selected enzymes on the soluble proteins in the latex were investigated from the changes of 16 kinds of amino acids, and the effect of deproteinization was evaluated by the change of the total amount of amino acids. The results show that the selected alkaline protease can hydrolyze the peptide bond of the soluble protein in the latex, and the amino acid composition of the protein varies during the hydrolysis. The law of latex in soluble protein detection limit of 1. 5 mg / L (5 μL injection, S / N = 2), the results are not affected by changes in amino acid composition of the protein. Amino acid composition analysis of bovine serum albumin showed that the method is of high accuracy.
