目的:对4种多聚物包被的球囊导管局部基因转移系统的在体转基因效果进行对比评估。方法:用由相同多聚物包被、携带不同的基因载体,及由不同的多聚物包被、携带相同的基因载体的球囊导管系统,将报告基因分别导入兔腹主动脉下段。结果:除由脂质体—腺病毒(Ad5)载体组成的基因转移系统外,其他3种系统均实现了在体基因转移。其中,由左旋多聚赖氨酸包被球囊导管—重组腺病毒载体(Ad5/LacZ)基因转移系统的基因转移效果最佳。结论:左旋多聚赖氨酸包被球囊导管—Ad5/LacZ基因转移系统是一种新型的、有效可行的在体局部基因转移系统。
OBJECTIVE: To compare the in vivo transgene effects of four multimeric coated balloon catheter local gene transfer systems. Methods: The reporter gene was introduced into the lower part of rabbit abdominal aorta by balloon catheter system, which was coated with the same polymer, carrying different gene carriers and coated with different polymers and carrying the same gene carrier. Results: In addition to the gene transfer system consisting of the liposome-adenovirus (Ad5) vector, all three other systems achieved in vivo gene transfer. Among them, the best results were achieved by the gene transfer system of L-poly-lysine-coated balloon catheter-recombinant adenovirus vector (Ad5 / LacZ). CONCLUSION: L-poly-L-lysine coated balloon catheter-Ad5 / LacZ gene transfer system is a new and effective in vivo gene transfer system.
