目的:优选北青龙衣总多糖的脱蛋白工艺。方法:以蛋白质脱除率和总多糖保留率为指标,通过单因素试验和正交试验比较Sevage法、三氯乙酸法(TCA)、木瓜蛋白酶法、酶-Sevage联用法及酶-TCA联用法对北青龙衣总多糖的脱蛋白质效果。结果:木瓜蛋白酶-TCA法除蛋白效果最好,最佳脱蛋白条件为木瓜蛋白酶用量3%,酶解时间2 h,酶解温度60℃,p H6,TCA质量分数6%。蛋白质脱除率84.76%,总多糖保留率82.48%。结论:不同脱蛋白质法对北青龙衣总多糖中蛋白质脱除效果有明显差异,酶-TCA法效果最佳且操作简便、耗时短。
Objective: To optimize the process of deproteinization of total polysaccharide of Beilong Qingyi. Methods: Sevage method, trichloroacetic acid method (TCA), papain method, enzyme-Sevage method and enzyme-TCA method were used to compare the protein removal rate and the total polysaccharide retention rate by single factor test and orthogonal test Deproteinization of total polysaccharide of Beilong Qingyi. Results: The best protein removal by papain-TCA was 3% papain, enzymatic hydrolysis time 2 h, hydrolysis temperature 60 ℃, p H6 and TCA 6%. The protein removal rate was 84.76% and the total polysaccharide retention rate was 82.48%. Conclusion: Different deproteinization methods have obvious effect on the removal of protein in total polysaccharide of Beilong Qingyi. Enzyme-TCA method is the best and simple in operation and short in time.
