目的:通过神经损伤评分(NSS)评分和体重测量,探讨回神颗粒减轻TBI的作用机制。方法:应用液压性脑损伤装置建立中度TBI模型,创伤压力为(170±10)k Pa,作用时间为(20±2)ms,随机将大鼠分为6组:正常组、假手术组、模型3 d组、模型7 d组、治疗3 d组、治疗7 d组。治疗3 d组和治疗7 d组于创伤后即刻用2 m L药液(含回神颗粒0.27 g)灌胃一次,此后再分别于每日早晚各灌胃一次,每次2 m L药液。分别于创伤后1 d、3d、7 d对正常组、假手术组、模型7 d组和治疗7 d组进行NSS量表评分;分别于创伤后1 d、3 d对模型3 d组和治疗3 d组进行NSS评分。分别于造模前和取材前称取每只动物的体重。观察NSS评分和体重的变化。结果:与正常组和假手术组比较,模型3 d组、模型7 d组、治疗3 d组、治疗7 d组各时间点NSS评分明显降低(P<0.01)。但与模型3 d组和模型7 d组的NSS评分相比较,治疗3 d组和治疗7 d组的NSS评分明显提高,具有显著性差异(P<0.01)。与正常组体重比较,模型组动物体重下降,具有明显差异(P<0.01);治疗组动物体重无明显下降(P>0.05)。比较每组动物造模前和取材前体重,发现模型组体重下降,具有明显差异(P<0.01);治疗组体重有所增加,具有明显差异(P<0.01)。结论:回神颗粒能明显改善TBI大鼠NSS评分,改善神经功能;促进大鼠的生长发育,减轻脑损伤。
OBJECTIVE: To investigate the mechanism of Yi Shen Granule in relieving TBI through the neurological injury score (NSS) score and body weight measurement. Methods: A moderate TBI model was established by using a hydraulic brain injury device. The traumatic stress was (170 ± 10) kPa and the duration of action was (20 ± 2) ms. Rats were randomly divided into 6 groups: normal group, sham operation group , Model 3 d group, model 7 d group, treatment group 3 d, treatment group 7 d. After 3 days of treatment and 7 days of treatment, intragastric administration of 2 m L of drug solution (containing Shenqian granules 0.27 g) was given immediately after trauma, and then 2 mg of liquid medicine . The NSS scale was used to evaluate the NSS scale on the 1st, 3rd, and 7th day after trauma in the normal group, the sham operation group, the model 7 d group and the 7 d treatment group respectively. 3 d group NSS score. Weigh the weight of each animal before modeling and before harvesting. NSS score and weight changes were observed. Results: Compared with normal group and sham operation group, the NSS score of model 3 d group, model 7 d group, treatment 3 d group and treatment 7 d group were significantly decreased at each time point (P <0.01). However, compared with the NSS scores of model group 3 d and model group 7 d, the NSS score of 3 d group and 7 d group increased significantly, with significant difference (P <0.01). Compared with the normal group, the body weight of the model group decreased significantly (P <0.01), while the body weight of the treated group had no significant difference (P> 0.05). The body weight of the model group was significantly lower than that of the model group (P <0.01). The body weight of the treatment group increased with significant difference (P <0.01). Conclusion: Shenxue granule can significantly improve the NSS score of TBI rats, improve neurological function, promote the growth and development of rats and relieve brain injury.
