目的研究局部使用外源性MGF-Ct24 E对雌性大鼠尿道损伤修复过程中内源性机械生长因子(MGF)基因表达的影响。方法将90只雌性未育Wistar大鼠随机分为3组,空白组(无损伤未注药)、对照组(损伤后注射生理盐水)、实验组(损伤后注射MGF-Ct24 E),通过模拟产伤制作尿道损伤模型,损伤后分别于尿道中段局部注射生理盐水、MGF-Ct24E,三组均于第1、4、7、10、14日(6只/日/组)截取中段尿道行SYBR Green RT-PCR半定量分析其内源性MGF mRNA基因的表达。结果空白组大鼠损伤后不同时间MGF mRNA表达差异无统计学意义(P>0.05);对照组MGFmRNA基因在第4、7、10日的表达量与空白组相比差异有统计学意义(P<0.05);与对照组相比,实验组MGF mRNA表达量在损伤后第1、4、7、10日明显增高(P<0.05);对照组MGF mRNA表达在损伤后第7日达到最高,实验组表达量在损伤后第4日达峰,实验组第14日MGF mRNA表达量较对照组略有增高,但差异无统计学意义。结论局部使用MGF-Ct24E可促进尿道损伤后内源性MGF mRNA表达并使其高峰提前。
Objective To investigate the effect of local administration of exogenous MGF-Ct24 E on the expression of endogenous mechanical growth factor (MGF) gene during urethral injury repair in female rats. Methods Totally 90 female Wistar rats were randomly divided into 3 groups: blank group (without injection); control group (saline injected after injury); experimental group (MGF-Ct24 E injected after injury) The injured urethral injury model was established. After injury, local injection of normal saline in the middle of the urethra, MGF-Ct24E, were performed on the 1st, 4th, 7th, 10th and 14th days (6 / day / Green RT-PCR semi-quantitative analysis of its endogenous MGF mRNA gene expression. Results There was no significant difference in the expression of MGF mRNA at different time points between the blank group and the blank group (P> 0.05). The expression of MGF mRNA in the control group on the 4th, 7th and 10th days was significantly lower than that of the blank group (P <0.05). Compared with the control group, the expression of MGF mRNA in the experimental group was significantly increased on the 1st, 4th, 7th and 10th days after injury (P <0.05). The MGF mRNA expression in the control group reached the highest on the 7th day after injury, The expression of MGF mRNA in the experimental group reached a peak on the fourth day after injury, and the MGF mRNA expression in the experimental group on the 14th day was slightly higher than that of the control group, but the difference was not statistically significant. Conclusion Local application of MGF-Ct24E can promote the expression of endogenous MGF mRNA after urethral injury and make its peak advance.
