目的分析重组SARS-CoVN蛋白的表达,并予以纯化。方法SDS-PAGE分析N蛋白的表达,于自动蛋白层析系统上,用His·BindTM柱亲和层析纯化。结果重组表达的N蛋白存在于裂解细菌上清中,分子量大小约49000u,N蛋白经亲和层析获得纯化,纯化蛋白能与病人血清反应。结论表明自动层析获得了纯化良好抗原性的N蛋白。
Objective To analyze the expression of recombinant SARS-CoVN protein and purify it. Methods The expression of N protein was analyzed by SDS-PAGE and purified on an automated protein chromatography system using His BindTM affinity chromatography. Results The recombinant N protein was found in the supernatant of lytic bacteria. The molecular weight of the N protein was about 49000u. The N protein was purified by affinity chromatography. The purified protein reacted with the serum of patients. The conclusion shows that purified and antigenically good N protein was obtained by automated chromatography.
