目的:探讨温肾补虚方对甲减大鼠下丘脑-垂体-甲状腺(HPT)轴干预的疗效性和量效性。方法:60只雄性SD大鼠,适应性饲养1周后,随机选取10只为正常组,余为实验组;实验组予丙硫氧嘧啶片(PTU)药品溶液(蒸馏水配成0.05%浓度)灌胃,持续造模4周后随机分为温低组、温中组、温高组、西药组、模型组,每组10只。分别予相应药物灌胃,周期28d。实验期间观察各组大鼠一般情况;干预后,检测各组大鼠血清TRH、TSH、FT4、FT3、TT4、TT3水平并观察甲状腺组织病理形态学改变。结果:与正常组比较,模型组大鼠饮食量、饮水量、体质量、肛温均下降(P<0.01),TRH、FT4、FT3、TT4、TT3下降(P<0.01),TSH上升(P<0.01),甲状腺组织损害严重。与模型组比较,温高组、西药组饮食量增加(P<0.01,P<0.05);温高组体质量增加(P<0.05,P<0.01);温高组、温中组肛温升高(P<0.05,P<0.01)。各治疗组大鼠血清TRH、FT4、FT3、TT4、TT3均升高(P<0.01,P<0.05),TSH下降(P<0.01,P<0.05)。HE染色发现各治疗组甲状腺腺泡均呈结节状增生,间质见充血、出血,与模型组比较,温中组腺泡增生及充血缓解更为明显。结论:温肾补虚方能有效改善甲减大鼠HPT轴功能,且存在一定量效性。
Objective: To investigate the effect of Wenshen Buxu Recipe on the hypothalamic-pituitary-thyroid (HPT) axis intervention in rats with hypothyroidism. Methods: Sixty male Sprague-Dawley rats were randomly selected for one week after the adaptive feeding. Ten rats were randomly selected as the normal group and the rest as the experimental group. The experimental group was given the drug solution of PTU (distilled water with 0.05% concentration) Gavage, continuous modeling 4 weeks later were randomly divided into warm low group, warm group, warm high group, western medicine group, model group, each group of 10. Respectively to the corresponding drug gavage, cycle 28d. During the experiment, the general condition of rats in each group was observed. After the intervention, the levels of serum TRH, TSH, FT4, FT3, TT4 and TT3 in each group were detected and the histopathological changes of thyroid tissue were observed. Results: Compared with the normal group, the dietary intake, water intake, body weight and rectal temperature in the model group decreased (P <0.01), the levels of TRH, FT4, FT3, TT4 and TT3 decreased <0.01), thyroid tissue damage is serious. Compared with the model group, the dietary intake of Wengao group and the western medicine group increased (P <0.01, P <0.05), the body weight of the Wistar group increased (P <0.05, P <0.01) High (P <0.05, P <0.01). The serum levels of TRH, FT4, FT3, TT4 and TT3 in rats of each treatment group were significantly increased (P <0.01, P <0.05), and TSH decreased (P <0.01, P <0.05). Hematoxylin-eosin staining showed that thyroid acinar showed nodular hyperplasia and hyperemia and hemorrhage in each treatment group. Compared with the model group, acinar hyperplasia and hyperemia in the treatment group were more obvious. Conclusion: Wenshen tonic prescription can effectively improve hypothyroid rat HPT axis function, and there is a certain amount of efficacy.
