目的 观察白介素-6(IL-6)对健康人中性粒细胞凋亡的抑制作用,及异丙酚对IL-6所致中性粒细胞凋亡抑制的影响。方法 分离20例健康志愿者的中性粒细胞,每例分成6部分,在RPMI1640培养液中孵育,其中1份作为对照,其余5份分别加入IL6、异丙酚、异丙酚的溶剂Intrlipid、IL-6+异丙酚、IL-6+Intrlipid,置入CO2培养箱中孵育24h,观察中性粒细胞的凋亡率并进行比较。结果IL-6可导致中性粒细胞凋亡的抑制(P<0.01);异丙酚和其溶剂Intralipid对健康人中性粒细胞的凋亡没有影响;与IL-6共同孵育时,异丙酚可部分逆转IL-6所导致的中性粒细胞凋亡障碍,而Intralipid则没有这种作用。结论 异丙酚可部分逆转IL-6导致的中性粒细胞凋亡障碍,提示异丙酚对细胞因子介导的全身炎症反应具有抑制作用。
Objective To investigate the inhibitory effect of interleukin-6 (IL-6) on the apoptosis of neutrophils and the effect of propofol on the apoptosis of neutrophils induced by IL-6. Methods Twenty healthy volunteers were divided into six groups, each of which was divided into six parts and incubated in RPMI1640 culture medium. One of them was used as a control. The other five were added with IL-6, propofol and propofol Solvent Intrlipid respectively. IL-6 + propofol, IL-6 + Intrlipid, incubated in CO2 incubator 24h, observed the neutrophil apoptosis rate and compared. Results IL-6 could inhibit the apoptosis of neutrophils (P <0.01). Propofol and its solvent Intralipid had no effect on the apoptosis of neutrophils. When incubated with IL-6, Phenol partially reversed IL-6-induced neutrophil apoptosis, whereas Intralipid did not. Conclusion Propofol can partially reverse the apoptosis of neutrophils induced by IL-6, suggesting that propofol has an inhibitory effect on cytokine-mediated systemic inflammatory response.
