转HJC-1和G6-EPSPS基因大米对五指山小型猪免疫指标的影响

目的探讨转HJC-1和G6-EPSPS基因抗虫耐草甘膦大米对五指山小型猪免疫指标的影响,观察其是否具有免疫毒性和潜在致敏性。方法分别用含70%转HJC-1和G6-EPSPS基因大米饲料、含70%9746亲本大米饲料、常规基础饲料,连续喂饲五指山小型猪30 d,观察动物一般行为表现、过敏症状和死亡情况,记录体重,计算免疫器官脏器系数,测定实验前后血清免疫指标、粪便胰蛋白酶和糜蛋白酶含量以及肠组织组胺水平。结果实验期间各组动物活动自如,未见腹泻等症状,未出现生病或死亡,体重和免疫器官脏器系数无异常改变。与对照组相比,转基因组实验前后血清IgG、IgE抗体、组胺及细胞因子IL-2、IFN-γ、IL-4、IL-10均无显著变化,亲本组除实验后血清组胺和IFN-γ高于对照组(P<0.05)外,其余指标未见改变。与对照组相比,转基因组和亲本组肠组织组胺水平及实验前后粪便胰蛋白酶和糜蛋白酶含量均未见显著变化。结论转HJC-1和G6-EPSPS基因大米及其亲本9746大米分别掺入饲料喂饲小型猪30 d,未见其对小型猪免疫指标产生不良影响。 OBJECTIVE: To investigate the effects of transgenic HGC-1 and G6-EPSPS-resistant insect-tolerant glyphosate-tolerant rice on the immune index of Wuzhishan mini-pigs and to observe whether they are immunotoxic and potentially allergenic. Methods Wuji Mountain mini-pigs were fed with 70% HJC-1 and G6-EPSPS gene rice, 70% 9746 parental rice and conventional basal diet for 30 days. The general behavior, allergic symptoms and death of animals , Body weight was recorded to calculate the organ coefficient of immune organs, serum immunological parameters were measured before and after the experiment, the contents of trypsin and chymotrypsin in the intestine, and histamine levels in intestinal tissue. Results During the experiment, animals in each group were free to move freely. No symptoms such as diarrhea occurred. No illness or death occurred. No abnormal changes were found in body weight and immune organs’ organ coefficients. Compared with the control group, serum IgG, IgE antibody, histamine and cytokines IL-2, IFN-γ, IL-4 and IL-10 had no significant changes before and after the transgene group experiment. And IFN-γ higher than the control group (P <0.05), the other indicators have not changed. Compared with the control group, there was no significant change in the histamine levels in the gut tissue of the transgenic and parental groups and the contents of trypsin and chymotrypsin in the feces before and after the experiment. Conclusion The transgenic HJC-1 and G6-EPSPS gene rice and its parent 9746 rice were fed with feed for 30 days, respectively, and no adverse effects on immunity index of mini-pigs were found.
