地胆草(Elephantopus scaber)的根(500g)经己烷脱脂,继之用乙醇提取。乙醇提取液在减压下蒸发干燥后,以300ml的蒸馏水制成糖浆状液体,再用氯仿提取。氯仿提取液经硅胶柱层析(用梯度为氯仿-己烷、氯仿、氯仿-甲醇的洗液洗脱),用制备型TLC对最初氯仿-甲醇组分进行纯化,分离得到化合物4,水组分经硅胶柱层析,用氯仿-甲醇-水(13:7:2)混合物的有机层洗脱,收集到分别为20ml的50个组分(根据TLC判断它们的均一性进行分组)。在硅胶板上对每一组分进行制备型TLC层析(氯仿-甲醇15:5作为流动相),从适当的组分中精制化合物1,2,3,分析它们的光谱和化
Roots of Elephantopus scaber (500 g) were defatted with hexane, followed by extraction with ethanol. The ethanol extract was evaporated to dryness under reduced pressure and made into a syrupy liquid with 300 ml of distilled water and extracted with chloroform. The chloroform extract was subjected to silica gel column chromatography (eluting with a gradient of chloroform-hexane, chloroform, and chloroform-methanol washings), and the initial chloroform-methanol fraction was purified by preparative TLC. The compound 4 was isolated and the water group was separated. The fractions were separated by silica gel column chromatography and eluted with an organic layer of a mixture of chloroform-methanol-water (13:7:2), and 50 fractions (20 ml each) were collected (grouped by their homogeneity as judged by TLC). Each component was subjected to preparative TLC chromatography on silica gel plates (chloroform-methanol 15:5 as mobile phase), and compounds 1 ,2,3 were purified from the appropriate components and analyzed for their spectra and chemistries.
