目的探讨复乳法制备丝裂霉素C(MMC)脂质体的工艺方法和包封率测定。方法采用复乳法制备MMC脂质体,用透射电镜观察所制备的MMC脂质体的形态,选择MMC的吸收波长,葡聚糖凝胶柱分离游离MMC和MMC脂质体,测定脂质体混悬液中的总药物量与游离药物量,完成MMC脂质体包封率的测定。结果所制得的MMC脂质体呈球形与类球形,未过柱除去游离药物的MMC脂质体混悬液中MMC的浓度为341.16μg/mL,用柱层析分离除去游离药物后所得的MMC脂质体定容至一定体积,其中药物浓度为1.41μg/mL,MMC脂质体平均包封率为8.25%。结论复乳法制得的脂质体的包封率较高,而且方法简单易行,药物产率也较高。
Objective To investigate the technology and encapsulation efficiency of mitomycin C (MMC) liposome prepared by double emulsion method. Methods MMC liposomes were prepared by double emulsion method. The morphology of MMC liposomes was observed by transmission electron microscopy. The absorption wavelength of MMC was selected. The free MMC and MMC liposomes were separated by Sephadex LH - The total amount of drug in the suspension and the amount of free drug were measured to determine the encapsulation efficiency of the MMC liposomes. Results The prepared MMC liposomes were spherical and spheroidized. The concentration of MMC in MMC liposome suspension without removing the free drug was 341.16μg / mL. The obtained MMC MMC liposome volume to a certain volume, which drug concentration was 1.41μg / mL, MMC liposome encapsulation efficiency was 8.25%. Conclusion The encapsulation efficiency of liposomes prepared by double emulsion method is high, and the method is simple and easy, and the drug yield is also high.
