目的:观察消癌舒(XAS)的抗炎镇痛作用并分析其作用机制。方法:昆明种小鼠随机分为模型组,阿司匹林组(0.2 g.kg-1),XAS低、中、高剂量组(0.375,0.75,1.5 g.kg-1),连续灌胃给药7 d。采用二甲苯致小鼠耳廓肿胀及角叉菜胶致小鼠足肿胀实验评价XAS的抗炎作用;采用冰醋酸致痛法及热板致痛法评价XAS的镇痛作用;采用Real time RT-PCR法测定炎性组织中肿瘤坏死因子α(TNF-α)及白介素1β(IL-1β)mRNA的表达。结果:XAS能明显抑制小鼠耳廓肿胀及足肿胀程度(P<0.05);对抗热板及醋酸刺激所致的小鼠疼痛反应(P<0.05);降低炎症组织中TNF-α及IL-1βmRNA的表达(P<0.05)。结论:XAS具有显著的抗炎、镇痛作用,其作用机制与抑制炎性组织中TNF-α,IL-1βmRNA的表达有关。
Objective: To observe the anti-inflammatory and analgesic effects of Xiao-Hua-Shu (XAS) and to analyze its mechanism of action. Methods: Kunming mice were randomly divided into model group, aspirin group (0.2 g.kg-1), XAS low, medium and high dose groups (0.375,0.75,1.5 g.kg-1) d. The anti-inflammatory effect of XAS was evaluated by xylene-induced mouse ear swelling and carrageenan-induced foot swelling in mice. The analgesic effect of XAS was evaluated by acetic acid induced pain and hot plate induced pain. Real time RT The expression of tumor necrosis factor α (TNF-α) and interleukin-1β (IL-1β) mRNA in inflammatory tissues were detected by PCR-PCR. Results: XAS could significantly inhibit the auricle swelling and foot swelling in mice (P <0.05), and alleviate the pain response in mice induced by hot plate and acetic acid stimulation (P <0.05), decrease the levels of TNF-α and IL- 1β mRNA expression (P <0.05). Conclusion: XAS has significant anti-inflammatory and analgesic effects, and its mechanism is related to the inhibition of the expression of TNF-α and IL-1βmRNA in inflammatory tissues.
