目的探讨纤溶酶原激活抑制物-1(PAI-1)基因4G/5G多态性与儿童青少年血糖及其相关指标的关系。方法选取北京市3919名7~18岁儿童青少年进行身体测量和FPG水平检测,部分样本检测FIns水平。利用基质支持的激光释放/电离飞行时间质谱分析(MALDI-TDF MS)对PAI-1基因4G/5G多态性进行检测。采用多元线性回归和Logistic二元回归分析基因多态性与血糖及相关指标的关系。结果校正研究人群、年龄和性别后发现,与5G5G基因型比较,4G等位基因携带者FPG水平较高(β=0.064,SE=0.029,P=0.025)。4G/5G多态性与IFG有相关性(OR=1.640,95%CI:1.166~2.309,P=0.005)。校正BMI后该多态性与IFG的相关性没有变化(FPG:β=0.063,SE=0.029,P=0.029;IFG:OR=1.628,95%CI:1.155~2.293,P=0.005)。结论 PAI基因4G/5G多态性与儿童青少年血糖水平和IFG有相关性,其效应独立于BMI的作用。
Objective To investigate the relationship between 4G / 5G polymorphism of plasminogen activator inhibitor-1 (PAI-1) gene and blood glucose in children and adolescents and their related indicators. Methods A total of 3919 adolescents aged 7-18 years in Beijing were selected for body measurement and FPG test. Some samples were tested for FIns level. PAI-1 gene 4G / 5G polymorphism was detected by matrix-supported laser emission / ionization time of flight mass spectrometry (MALDI-TDF MS). Multivariate linear regression and Logistic regression analysis of the relationship between gene polymorphism and blood glucose and related indicators. Results Correction of the study population, age and gender found that patients with the 4G allele had higher levels of FPG compared with the 5G5G genotype (β = 0.064, SE = 0.029, P = 0.025). The 4G / 5G polymorphism was associated with IFG (OR = 1.640, 95% CI: 1.166-2.309, P = 0.005). There was no change in the association of this polymorphism with IFG after adjustment for BMI (FPG: β = 0.063, SE = 0.029, P = 0.029; IFG: OR = 1.628,95% CI: 1.155-2.293, P = 0.005). Conclusion The 4G / 5G polymorphism of PAI gene is correlated with the level of plasma glucose and IFG in children and adolescents, and its effect is independent of BMI.
