目的:探讨透骨消痛胶囊含药血清影响软骨细胞线粒体凋亡通路的作用机制。方法:SD大鼠膝关节软骨建立体外培养的软骨细胞,Ⅱ型胶原原位杂交鉴定。第3代软骨细胞培养72h,SNP诱导软骨细胞凋亡,采用含药血清干预凋亡软骨细胞,Hocehst 33342染色检测软骨细胞的凋亡率,Real time RT-PCR检测Bax、Bcl-2、Caspase-9、Caspase-3 mRNA表达。结果:干预24h,软骨细胞的OD值,对照组、实验2组、实验3组明显高于空白组(P<0.05);软骨细胞Bcl-2mRNA表达,对照组、实验2组、实验3组明显高于空白组(P<0.05);软骨细胞Bax、Caspase-9、Caspase-3 mRNA表达,对照组、实验2组、实验3组明显低于空白组(P<0.05)。结论:透骨消痛胶囊含药血清能下调软骨细胞促凋亡基因Bax、Caspase-9、Caspase-3表达,上调抗凋亡基因Bcl-2表达,抑制软骨细胞线粒体凋亡通路的信号转导。
Objective: To investigate the mechanism of the effect of the traditional Chinese medicine of the traditional Chinese medicine TouJiaoJiaoTong capsule on mitochondrial apoptosis pathway of chondrocytes. Methods: Chondrocytes cultured in vitro were cultured in articular cartilage of SD rats, and type Ⅱ collagen was identified by in situ hybridization. The chondrocytes of the 3rd generation were cultured for 72h. The apoptosis of chondrocytes was induced by SNP. The apoptotic rate of chondrocytes was detected by Hoechst 33342 staining and the expression of Bax, Bcl-2 and Caspase- 9, Caspase-3 mRNA expression. Results: The OD values of chondrocytes in control group, experimental group 2 and experimental group 3 were significantly higher than those in blank group (P <0.05) after intervention for 24 h. The expression of Bcl-2 mRNA in chondrocytes in control group, experimental group 2 and experimental group 3 were significantly (P <0.05). The expressions of Bax, Caspase-9 and Caspase-3 mRNA in chondrocytes were significantly lower in control group, experimental group 2 and experimental group 3 than those in blank group (P <0.05). CONCLUSION: The traditional Chinese medicine can decrease the expression of Bax, Caspase-9, Caspase-3 and up-regulate the expression of anti-apoptotic gene Bcl-2 and inhibit the signal transduction of mitochondrial apoptosis pathway in chondrocytes .
