目的探讨没食子酸(gallic acid,GA)体外对胃癌细胞株MGC-803的抑制作用及机制。方法体外培养人胃癌细胞株MGC-803,MTT法观察细胞生长情况;Hoechst-33258、Annexin V-FITC/PI检测细胞凋亡情况;RT-PCR方法研究Survivin的mRNA的变化。结果 MTT检测6.25~50μmol/LGA可抑制MGC-803的生长,并呈剂量依赖性。Hoechest-33258显示出明显的细胞凋亡形态。用Annexin V-FITC/PI检测细胞凋亡率有显著的剂量依赖性。同时RT-PCR反映了GA作用后,胃癌细胞中Survivin mRNA表达减少。结论 GA可抑制胃癌细胞MGC-803的增殖,其作用机制可能与下调相关凋亡基因Survivin有关。
Objective To investigate the inhibitory effect of gallic acid (GA) on gastric cancer cell line MGC-803 and its mechanism. Methods The human gastric cancer cell line MGC-803 was cultured in vitro. The cell growth was observed by MTT assay. The apoptosis of cells was detected by Hoechst-33258 and Annexin V-FITC / PI. The mRNA expression of Survivin was detected by RT-PCR. Results The MTT assay of 6.25 ~ 50μmol / LGA inhibited the growth of MGC-803 in a dose-dependent manner. Hoechest-33258 showed a distinct apoptotic morphology. Annexin V-FITC / PI detection of apoptosis rate was significantly dose-dependent. At the same time RT-PCR reflects the role of GA, gastric cancer cells Survivin mRNA expression decreased. Conclusion GA can inhibit the proliferation of gastric cancer cell line MGC-803, which may be related to the down-regulation of Survivin.
