目的:观察丹参毛状根对离体大鼠心肌缺血再灌注损伤的保护作用。方法:采用Langendorff离体心脏恒压灌流方法,以0.5 mL.min-1的灌流速度,生药质量浓度分别为14,28,56 mg.L-1的丹参毛状根,与k-H灌流液同时灌流心脏10min,然后全心停灌30 min,复灌60 min。结果:离体实验发现,丹参毛状根能够增加冠脉流量,不但降低心律失常的发生率,而且推迟心律失常的发生时间,缩短持续时间和恢复时间,减低LDH,MDA的释放,增加SOD活性。结论:丹参毛状根对大鼠离体心脏缺血再灌注损伤具有一定的保护作用;与丹参药材相比无显著性差异。
Objective: To observe the protective effect of hairy root of Salvia miltiorrhiza on myocardial ischemia-reperfusion injury in isolated rat hearts. METHODS: A Langendorff isolated heart was used as a constant pressure perfusion method. With a perfusion rate of 0.5 mL.min-1, the hairy roots of Salvia miltiorrhiza with a crude drug concentration of 14, 28, 56 mg.L-1 were perfused simultaneously with the perfusate of kH. The heart was left for 10 minutes, then the heart was stopped for 30 minutes and re-instilled for 60 minutes. Results: In vitro experiments showed that hairy roots of Salvia miltiorrhiza can increase coronary flow, not only reduce the incidence of arrhythmia, but also delay the occurrence of arrhythmia, shorten the duration of the arrhythmia, shorten the duration and recovery time, reduce the release of LDH, MDA, and increase the activity of SOD. . Conclusion: Hairy root of Salvia miltiorrhiza has a protective effect on rat heart ischemia-reperfusion injury. There is no significant difference compared with Salvia miltiorrhiza.
