目的探讨泽黄颗粒对糖尿病大鼠肾组织nephrin表达的调节。方法用高脂高糖饲料加小剂量STZ诱导糖尿病大鼠模型,随机分为糖尿病模型组(DM组)、泽黄颗粒治疗组(Z组)、格列吡嗪对照组(G组)和正常对照组(N组)。实验第12周末检测BG、Cc(r肌酐清除率)及UAER(尿白蛋白排泄率),观察肾组织病理改变,运用免疫组织化学、Western-Blot、Real-Time PCR法检测肾脏nephrin蛋白及mRNA表达的变化。结果泽黄颗粒可以改善DM大鼠Ccr,降低UAER,改善肾脏病理,减轻足突融合,维持足细胞相关蛋白分子的分布与表达。结论泽黄颗粒通过上调足细胞相关蛋白分子nephrin水平而减轻足细胞损伤,对糖尿病大鼠足细胞损伤有保护作用。
Objective To investigate the effect of Zehuang Granule on the regulation of nephrin expression in kidney of diabetic rats. Methods Diabetic rat models were induced by high fat, high glucose diet and low dose STZ. They were randomly divided into diabetic model group (DM group), Zehuang granule treatment group (Z group), glipizide control group (G group) and normal group. Control group (N group). At the end of the experiment, BG, Cc (r creatinine clearance) and UAER (urinary albumin excretion rate) were measured at the end of the experiment to observe the pathological changes of the kidney. The nephrin protein and mRNA of the kidney were detected by immunohistochemistry, Western-Blot and Real-Time PCR. Change in expression. Results Ze Huang Granule can improve Ccr in DM rats, reduce UAER, improve renal pathology, reduce foot process fusion, and maintain the distribution and expression of podocyte-associated protein molecules. Conclusion Zehuang granules can reduce podocyte injury by up-regulating the nephrin level of podocyte-associated protein molecules, and have protective effects on podocyte injury in diabetic rats.
