目的 :探讨宫颈鳞癌组织中EphA2的表达。方法 :应用免疫组化法观察EphA2在 2 0 6例FIGOⅠa Ⅳb期的宫颈鳞癌组织中的表达 ,从中选择 2 0例不同EphA2蛋白表达的病例 ,应用激光捕获显微切割技术 (Lasercapturemicrodissection ,LCM)从冰冻组织切片中选切大约 5 0个宫颈鳞癌细胞 ,应用RT PCR方法检测EphA2mRNA的表达。结果 :2 0 6例宫颈鳞癌组织中 ,EphA2蛋白阴性表达 2 3例 (1 1 .2 % ) ,弱阳性表达 94例 (4 5 .6 % ) ,阳性表达 72例 (35 .0 % ) ,强阳性表达 1 7例 (8.3% ) ;EphA2蛋白与其mRNA表达水平不完全一致 ;EphA2蛋白表达与宫颈鳞癌患者的年龄、FIGO分期及淋巴结转移无关 ,总生存期在EphA2染色强度 0~ 3级之间差异有统计学意义 (P <0 .0 5 )。结论 :EphA2在宫颈鳞癌中的高表达预示患者的生存差。EphA2有望成为抗宫颈癌治疗的新靶点
Objective: To investigate the expression of EphA2 in cervical squamous cell carcinoma. Methods: The expression of EphA2 was detected by immunohistochemistry in 20 0 cases of FIGOⅠa Ⅳb cervical squamous cell carcinoma. Twenty cases of different EphA2 protein expression were selected. Laser capture microdissection (LCM) About 50 cervical squamous cell carcinoma cells were selected from frozen tissue sections and the expression of EphA2 mRNA was detected by RT-PCR. Results: The negative expression of EphA2 protein in 266 cases of cervical squamous cell carcinoma was 23 cases (112%), 94 cases (45.6%) had weakly positive expression, and 72 cases (35.0%) had positive expression of EphA2 protein , Strong positive expression in 17 cases (8.3%); EphA2 protein expression level is not exactly the same; EphA2 protein expression and cervical squamous cell carcinoma patients age, FIGO stage and lymph node metastasis, the overall survival of the EphA2 staining intensity 0-3 The difference between the two groups was statistically significant (P <0.05). Conclusion: The high expression of EphA2 in cervical squamous cell carcinoma predicts the poor survival of patients. EphA2 is expected to become a new target for the treatment of cervical cancer
