本研究组已证实,部分去传入猫脊髓背角组织提取液中存在某种促神经元存活的神经营养活性物质。为了探讨针刺对该活性物质含量及其对神经元存活的影响,作者采用5只双侧备用根猫(切除双侧Ll~L5,L7~S2背根节,保留L6为备用根),每日针刺一侧L6背神经支配范围内的两组穴位(足三里与悬钟、伏兔与三阴交)之一,术后5天分别取双侧T12~S2背角组织,匀浆、离心,取上溶液进行电泳和细胞培养。经聚丙烯酸胺梯度电泳后行光密度扫描,发现针刺侧相对迁移率(RM)0.11蛋白带的吸收峰面积百分比(11.78±l.08%)较非针刺侧着(10.12±0.83%)明显增高(P<0.05),表明针刺使RM0.11带蛋白质的含量增加。用本组建立的简易细胞钓蛋白带法将鸡胚背根节(DRG)的神经细胞悬液倒于电泳凝胶条上进行培养,发现凝胶条上RM0.11蛋白带所在区域内(RM0.10~0.13),有密集的神经细胞贴附。恒定面积由(1.02×104μm2/视野)内,针刺侧平均细胞数(19.09±2.12个)较非针刺侧者(8.64±1.12个)明显增多(P<0.05)。聚丙烯酸胺梯度电泳测定分子量后显示RM0.11带蛋白质分子量约为130KD左右。上述结果表明,部分去传入猫背角组织中分子量约130KD左右的蛋白质具有营养DRG神经元的作用,针刺可能通过该类?
