目的:探讨超氧化物歧化酶(SOD)对肝癌细胞增殖的抑制作用。方法:利用逆转录病毒载体介导的基因转移技术,将人铜锌SOD基因导入人肝癌细胞系HepG2细胞内,获得高表达。检测其对HepG2细胞生物学特性的影响。结果:与对照相比SOD基因转染后的肝癌细胞生长受到抑制,细胞体积变大,分裂象减少,S期细胞比率减少,软琼脂集落形成率低,裸鼠成瘤瘤体小。结论:铜锌SOD基因具有抑制HepG2肝癌细胞增殖的作用。
Objective: To investigate the inhibitory effect of superoxide dismutase (SOD) on proliferation of hepatoma cells. METHODS: Human copper-zinc SOD gene was introduced into human hepatocellular carcinoma cell line HepG2 cells using a retroviral vector-mediated gene transfer technique to obtain high expression. Its effect on the biological characteristics of HepG2 cells was examined. RESULTS: Compared with the control, the growth of hepatoma cells transfected with SOD gene was inhibited, the cell volume became larger, the mitotic pattern decreased, the ratio of S phase cells decreased, the formation rate of soft agar colonies was low, and the tumor formation of nude mice was small. Conclusion: Copper-zinc SOD gene can inhibit the proliferation of HepG2 cells.
