利用oct4、sox2、c-Myc、klf4基因转染体细胞可获得与胚胎干细胞(ESC)类似的诱导性多能干细胞。其中,Oct4作为体细胞重编程必不可少的核心转录因子,在转录调节、信号传导通路、表观遗传修饰等方面具有重要调控作用。本文通过阐述Oct4及其亚型调控ESC多能性的作用机制,揭示其在体细胞重编程中的调节作用,以期为研究Oct4对牙髓干细胞的调节作用提供一定的理论依据。
Induced pluripotent stem cells similar to embryonic stem cells (ESC) can be obtained by transfecting somatic cells with oct4, sox2, c-Myc and klf4 genes. Among them, Oct4, as an essential core transcription factor of somatic cell reprogramming, plays an important regulatory role in transcription regulation, signal transduction pathway and epigenetic modification. In this review, we elucidate the regulatory mechanism of Oct4 and its subtypes in the regulation of ESC pluripotency and reveal their regulatory roles in somatic cell reprogramming. The purpose of this study is to provide some theoretical evidences for the regulation of Oct4 on dental pulp stem cells.
