目的:观察附子理中汤对脾阳虚大鼠心房利尿钠肽(ANP)含量和非可溶性鸟苷酸环化酶(pGC)mRNA表达及胃肠动力学的影响。方法:清洁级Wistar大鼠100只随机分为对照组、模型组、附子理中汤低、中、高剂量组5组,每组20只。对照组常态饲养。模型组采用“肩胛骨间棕色脂肪组织(BAT)切除术+高脂饲料喂养+隔日寒冷环境刺激”方法造模。术后第1天喂高脂饲料共3周。在模型组基础上,附子理中汤低、中、高剂量组术后第1天按10,20,40 g·kg-1ig给药,对照组及模型组则给予等容量的生理盐水,每天1次,连续3周。3周后第1天测空肠肌活动,然后取结肠标本,ELISA法检测ANP含量,RT-PCR法检测pGC mRNA表达。结果:和对照组比较模型组的ANP含量、pGC mRNA相对表达量降低(P<0.05或P<0.01),慢波频率显著下降(P<0.01),快波出现频率、最大振幅、每丛快波数均增高(P<0.05或P<0.01);纵肌收缩力振幅和环肌收缩力振幅均显著增大(P<0.01)。和模型组比较,附子理中汤高剂量组的ANP含量、pGC mRNA相对表达量升高(P<0.05或P<0.01);附子理中汤中剂量组慢波频率上升(P<0.05),快波出现频率、最大振幅、每丛快波数均减小(P<0.05或P<0.01);附子理中汤高剂量组慢波频率显著上升(P<0.01),快波出现频率、最大振幅、每丛快波数均显著减小(P均<0.01);中剂量组纵肌收缩力振幅减小(P<0.05),高剂量组纵肌收缩力振幅和环肌收缩力振幅均显著减小(P<0.01)。结论:附子理中汤“止泻”的一种可能机制在于一方面上调ANP的含量及pGC mRNA表达量,进而恢复正常胃肠动力学,另一方面通过对水盐重吸收的调节作用而起效。
Objective: To observe the effects of Fuzi Lizhong Decoction on atrial natriuretic peptide (ANP) content, non-soluble guanylate cyclase (pGC) mRNA expression and gastrointestinal motility in rats with spleen yang deficiency. Methods: One hundred clean Wistar rats were randomly divided into control group, model group and Fuzi Lizhong Tang low, medium and high dose groups of 5, 20 rats in each group. Control group feeding normally. The model group was modeled using the technique of “shoulder-to-shoulder brown adiposectomy (BAT) resection + high-fat diet feeding + cold weather every other day”. The first day after the high-fat diet were fed for 3 weeks. The activities of jejunum were measured on the first day after 3 weeks. The colon samples were collected. The content of ANP was detected by ELISA. The expression of pGC mRNA was detected by RT-PCR. Results: Compared with the control group, the ANP content of the model group decreased the relative expression of pGC mRNA (P <0.05 or P <0.01) and the frequency of slow wave significantly decreased (P <0.01). The frequency and maximum amplitude of fast wave, Wave number increased (P <0.05 or P <0.01), amplitude of longitudinal muscle contractility and amplitude of myocardial contractility increased significantly (P <0.01). Compared with the model group, the ANP content and the relative expression of pGC mRNA in the high-dose Fuzi Lizhong decoction group increased (P <0.05 or P <0.01) The frequency of fast wave, the maximum amplitude and the number of fast wave per bundle decreased (P <0.05 or P <0.01), and the frequency of slow wave of high dose of Fuzi Li Zhong Tang significantly increased (P <0.01) (P <0.01). The amplitude of longitudinal muscle contractility decreased in the middle dose group (P <0.05), and the amplitude of longitudinal muscle contraction and amplitude of the contractile force in the high dose group decreased significantly (P <0.01). Conclusion: A possible mechanism of aconite in the soup “diarrhea ” is that on the one hand, the content of ANP and pGC mRNA expression, and then restore normal gastrointestinal motility, on the other hand through the regulation of water and salt reabsorption And effective.
