Objective:To study the role of hypermethylation of 5’ CpG island of p16 gene in arcinogenesis and progression of gynecologic malignancy. Methods: PCR-based analysis was performed to detect the hypermethylation of 5’ CpG island of exon 1 in p16 gene in ervical,endometrial and ovarian carcinomas and corresponding normal tissues. Results: ypermethylation of p16 gene was detected in 6 of 30 cervical, 8 of 34 endometrial, 4 of 30 ovarian carcinomas respectively. Hypermethylation of p16 gene in cervical and endornetrial carcinomas was significantly higher at late stage(Ⅲ-Ⅳ )and low grade than that at early stage(Ⅰ-Ⅱ )and high grade. Conclusions: Hypermethylation of pl6 gene may play an important role in carcinogenesis and progression of gynecologic malignancy.
