本实验采用噻唑蓝(MTT)比色法观察氧化型低密度脂蛋白(OLDL)对体外培养的牛主动脉平滑肌细胞增殖的影响及抗氧化剂丙丁酚的作用。结果表明OLDL能降低一氧化氮(NO)含量和刺激血管平滑肌细胞(SMC)增殖(P<0.05),丙丁酚能拮抗OLDL刺激SMC增殖作用,同时增加NO含量。NO合成酶抑制剂硝基-L-精氨酸增强OLDL刺激血管平滑肌细胞增殖作用,且部分抵消丙丁酚的桔抗作用。提示丙丁酚能抑制OLDL刺激血管平滑肌细胞增殖作用,且与其促进平滑肌细胞合成释放一氧化氮有关。
In this study, MTT assay was used to observe the effects of oxidized low density lipoprotein (OLDL) on the proliferation of cultured aortic smooth muscle cells in vitro and the effect of the antioxidant probucol. The results showed that OLDL can reduce the content of nitric oxide (NO) and stimulate the proliferation of vascular smooth muscle cells (SMC) (P <0.05). Probucol can antagonize the proliferation of SMC stimulated by OLDL and increase the content of NO. Nitric-L-arginine, an NO synthase inhibitor, enhances the proliferation of vascular smooth muscle cells stimulated by OLDL and partially counteracts the anti-citrullination effect of probucol. Probucol can inhibit OLDL stimulated the proliferation of vascular smooth muscle cells, and its promotion of smooth muscle cell synthesis and release of nitric oxide.
