抗原、抗体间可发生特异性免疫反应 ,将发光物质标记在抗体上 ,经免疫反应和化学发光反应 ,测量光信号可对待测抗原进行定性、定量分析。本实验以吖啶酯DMAE·NHS标记TSH单克隆抗体 ,建立了TSH化学发光免疫分析方法。该方法分析灵敏度为 0 .0 1mIU/L ,批内相对标准偏差为 4 .2 9%~ 6 71% ,批间相对标准偏差为 4 .36 %~ 8.14 % ;平均回收率为 98%。与TSHIRMA法的相关方程为Y =0 .0 2 +0 .92X ,相关系数为 0 .993;与CibaCorningTSHCLIA测量值的相关方程为Y =- 0 .16 +0 .92X ,相关系数为 0 .986。
Antigens, antibodies can occur between the specific immune response, the luminescent substances marked on the antibody, the immune response and chemiluminescence reaction, the measured optical signal can be tested antigen qualitative and quantitative analysis. In this experiment, TSH chemiluminescence immunoassay was established with acridine ester DMAE · NHS labeled TSH monoclonal antibody. The analytical sensitivity of this method was 0.1 lmIU / L, with a relative standard deviation of 4.29% ~ 671%. The relative standard deviation was 4.36% ~ 8.14%. The average recovery was 98%. The correlation equation with TSHIRMA method is Y = 0.002 + 0.92X, the correlation coefficient is 0.993; the correlation equation with the measured value of CibaCorningTSHCLIA is Y = -0.16 +0.92X, the correlation coefficient is 0.986 .
