目的对一种由海绵Spongosoritessp.提取的双吲哚碱bromotopsentin(BSM)的抗氧化及抗癌活性进行研究。方法通过DPPH、TEAC等一系列与抗氧化活性相关联的分析方法,分析了BSM的抗氧化活性,并通过细胞毒性和抗增殖实验分析BSM对人宫颈癌细胞(HeLa)及人肺癌细胞(A549)生长的抑制作用。结果BSM对稳定自由基1,1-二苯基苦基苯肼(DPPH)的清除活性超过抗坏血酸的相应活性,其IC50值为48.9μmol.L-1;用6-羟基-2,5,7,8-四甲基苯并二氢吡喃-2-羧酸(Trolox)等效抗氧化能力(TEAC)方法测定BSM总的抗氧化能力大约为1.2,高于抗坏血酸的1.04。BSM对HeLa细胞及A549细胞的生长具有抑制作用,其IC50分别为2.3、24.5μmol.L-1。结论BSM有良好的抗氧化能力,同时对HeLa细胞及A549细胞有较强的细胞毒性作用。
OBJECTIVE To study the antioxidant and anticancer activity of a novel dipotenoid bromotopsentin (BSM) extracted from Spongosoritessp. Methods The antioxidant activity of BSM was analyzed by a series of analytical methods such as DPPH and TEAC. The cytotoxicity and antiproliferation assay were used to analyze the effect of BSM on the proliferation of human cervical cancer cell line (HeLa) and human lung cancer cell lines (A549 ) Growth inhibition. Results The scavenging activity of BSM to scavenging free radical 1,1-diphenylcarbitol (DPPH) was higher than that of ascorbic acid with an IC50 value of 48.9 μmol·L-1. With 6-hydroxy-2,5,7 , 8-tetramethylchroman-2-carboxylic acid (Trolox) equivalent antioxidant capacity (TEAC) method to determine the total antioxidant capacity of BSM is about 1.2, higher than 1.04 ascorbic acid. BSM had an inhibitory effect on the growth of HeLa cells and A549 cells with IC50 of 2.3 and 24.5μmol.L-1, respectively. Conclusion BSM has a good antioxidant capacity, while HeLa cells and A549 cells have a strong cytotoxicity.
