采用三种不同生根法,成功地诱导梨砧木BP10030试管苗生根.试管生根法最适的吲哚丁酸(IBA)浓度为2mg·1~(-1),生根率、平均根数和根长度分别为95%、7.4和3.4cm.用2—8mg·l~(-1)的低浓度IBA浸枝处理,生根率均在90%以上,其中以4mg·l~(-1)的IBA促根效果最佳.用10—500mg·l~(-1)的高浓度IBA浸枝处理,生根较差.尽管试管生根法容易诱导试管苗形成不定根,但是不定根形成部位产生大量愈伤组织,试管苗移栽成活率低,初期生长缓慢.采用低浓度IBA浸枝生根法,不定根形成部位不产生愈伤组织,移栽成活率高,试管苗初期生长快.
