目的:观察前列汤对实验性慢性非细菌性前列腺炎(CAP)大鼠Th1/Th2细胞平衡的影响,探讨前列汤治疗慢性非细菌性前列腺的作用机理。方法:采用前列腺腹叶注射角叉菜角的方法,建立CAP大鼠模型。用前列汤按10mL/(kg.d)灌胃(C组),并与模型对照组(B组)及正常组(A组)作对照。分别在给药后第30天后,采用流式细胞术检测各组动物外周血CD3+、CD8-、T细胞内细胞因子干扰素γ(IFN-γ)和白细胞介素4(IL-4)的表达,分别代表Th1和Th2细胞,测定各自在淋巴细胞所占比例,计算Th1/Th2比值。结果:与A组相比,B、C组Th1细胞数均升高,Th1/Th2比值升高,差异有显著性(P<0.05),Th2细胞数差异无统计学意义(P>0.05);C组与B组相比,Th1细胞数下降,;Th1/Th2比值下降,差异有显著性(P<0.05)。结论:(1)模型组Th1/Th2比值明显升高,支持慢性非细菌性前列腺炎发病中存在Th1/Th2比值失衡学说。(2)前列汤选择性降Th1数量,逆转失衡的Th1/Th2比值是其治疗慢性非细菌性前列腺炎的重要机理之一。
OBJECTIVE: To observe the effect of Qianlie Tang on the balance of Th1/Th2 cells in experimental chronic abacterial prostatitis (CAP) rats and to explore the mechanism of Qianlie Decoction in treating chronic non-bacterial prostate. Methods: A rat model of CAP was established by intraperitoneal injection of carrageenan in the prostate gland. Forehead soup was intragastrically administered at 10 mL/(kg.d) (group C) and compared with model control group (group B) and normal group (group A). After 30 days after administration, the expression of cytokines interferon-γ (IFN-γ) and interleukin-4 (IL-4) in peripheral blood of CD3+, CD8-, and T cells in each group of animals was detected by flow cytometry. Representing Th1 and Th2 cells, respectively, determine the percentage of lymphocytes in each, and calculate the Th1/Th2 ratio. Results: Compared with group A, the number of Th1 cells in group B and group C increased, and Th1/Th2 ratio increased. The difference was significant (P<0.05). There was no significant difference in the number of Th2 cells (P>0.05). Compared with group B, the number of Th1 cells in group C decreased, and Th1/Th2 ratio decreased, the difference was significant (P<0.05). Conclusion: (1) The ratio of Th1/Th2 in the model group was significantly increased, supporting the existence of an imbalance between Th1/Th2 ratios in the pathogenesis of chronic nonbacterial prostatitis. (2) Qianlietang selectively reduces the number of Th1 and reverses the unbalanced Th1/Th2 ratio, which is one of the important mechanisms for the treatment of chronic abacterial prostatitis.
