目的 建立高低不同转移特性的人骨肉瘤克隆细胞株 .方法 通过克隆技术和细胞电泳 ,从我们建立的骨肉瘤细胞系 L TH- OS(简称 S)中获得两株细胞 S1 和 S2 ,测定两种细胞的增殖率 ,利用软琼脂克隆形成率和裸鼠体内成瘤实验 ,比较细胞的转移特性 .结果 S1 的细胞电泳率高达 1.0 ,S2 最低为 0 .70 ,S1 的群体倍增时间为 30 .8h,S2 为 45 .0 h.克隆形成率分别为 5 .2 %和 2 .1% ,裸鼠右后肢皮下接种 S1 和 S2 发现S1 成瘤快 ,且瘤体大 ,原位接种时 S1 出现广泛的肺转移 .结论 S1 和 S2 是从 S母细胞中分离出的具有不同转移特性的细胞株
Objective To establish human osteosarcoma clonal cell line with different metastatic characteristics.Methods Two cell lines, S1 and S2, were obtained from our established osteosarcoma cell line L TH-OS (S) by cloning technology and cell electrophoresis, The rate of cell migration was compared with the rate of soft agar colony formation and tumor formation in nude mice.Results The cell electrophoresis rate of S1 was up to 1.0, the minimum of S2 was 0.70, the population doubling time of S1 was 30.8 h, S2 was 45 .0 h. The rate of colony formation was 5.2% and 2.1% respectively. S1 and S2 were subcutaneously inoculated into the right hind limb of nude mice, Lung metastasis.Conclusion S1 and S2 are cell lines with different metastatic characteristics isolated from S blasts
