目的:采用RAPD技术对湖南境内的甘薯品种进行多样性分析,并与境外的部份品种进行比较分类与鉴定研究。方法:从100个10bp的随机引物中筛选出20个扩增效果较好的引物进行扩增。结果:共产生188条带,其中118条为多态性带,占62.8%,平均每个引物扩增出的DNA多态性带数为5.90条,根据扩增结果构建出反映品种多样的UPGMA聚状图,31个品种可以划分为4大类。结论:从分子水平揭示了湖南境内部分甘薯品种的亲缘关系。
OBJECTIVE: To analyze the diversity of sweet potato varieties in Hunan Province by RAPD technique and to compare and identify them with some overseas cultivars. Methods: 20 primers with good amplification effect were selected from 100 10bp random primers for amplification. Results: A total of 188 bands were produced, of which 118 were polymorphic bands, accounting for 62.8%. The average number of DNA polymorphic bands amplified by each primer was 5.90. Based on the amplified results, UPGMA Poly graph, 31 varieties can be divided into four categories. Conclusion: The molecular relationship between some sweet potato varieties in Hunan Province was revealed.
