目的 研究白细胞介素 1β(interleukin 1β,IL 1β)对视网膜组织中核因子 κB(nuclearfactor κB ,NF κB)活化的诱导作用 ,以及四氢化吡咯 二硫代氨基甲酸酯 (pyrrolidinedithiocarbamate,PDTC)对NF κB的抑制作用。方法 将 16只Sprague Dawley (SD)大鼠随机等分为A、B两组。A组实验眼 (右眼 )玻璃体腔内 (下同 )注入 5 μl的IL 1β(5× 10 7U/L) ,对照眼 (左眼 )注入等量PSS ;B组实验眼先注入 10 μl的PDTC液 (1mmol/L) ,对照眼注入等量PSS ,1h后双眼再分别注入5 μl的IL 1β(5× 10 7U/L) ;均分别于 4h和 2 4h后处死大鼠 ,摘除眼球 ,取视网膜组织制备核提取物 ;用泳动迁移试验检测两组视网膜中NF κB/DNA结合活性。结果 SD大鼠玻璃体腔内注药 4h后 ,A组实验眼NF κB/DNA结合活性较对照眼增加 ,B组实验眼NF κB/DNA结合活性较对照眼降低 ;2 4h后 ,A组实验眼及B组对照眼的NF κB/DNA结合活性均较 4h者降低。结论 视网膜NF κB可被眼内IL 1β激活 ,NF κB在视网膜炎性反应中可能起重要调控作用
Objective To investigate the effect of interleukin 1β (IL 1β) on the activation of nuclear factor κB (NF κB) in retina and the effect of pyrrolidinedithiocarbamate (PDTC) on NF κB Inhibition. Methods Sixteen Sprague Dawley (SD) rats were randomly divided into A and B groups. A group of experimental eye (right eye) intravitreal (same below) into the 5 l of IL 1β (5 × 10 7U / L), the control eye (left eye) into the same amount of PSS; PDTC solution (1 mmol / L) was injected into the same volume of eyes in the same volume of PSS. After 1h, both eyes were injected with 5μl of IL-1β (5 × 10 7U / L). Rats were sacrificed at 4h and 24h, The retina was used to prepare the nuclear extracts. The NF κB / DNA binding activity in the retina was detected by the migration assay. Results After 4h injection into the vitreous of SD rats, the NF κB / DNA binding activity in experimental group A was higher than that in control group. The NF κB / DNA binding activity in experimental group B was lower than that in control group. After 24 hours, And the NF κB / DNA binding activity of B group was lower than that of 4h. Conclusions Retinal NF κB can be activated by IL 1β in the eye, and NF κB may play an important regulatory role in retinal inflammatory reaction
