目的 研究6%羟乙基淀粉(贺斯,200/0.5)血液稀释对大鼠全脑缺血/再灌注期血管内皮细胞细胞间粘附分子-1(ICAM-1)表达的影响。方法84只Wistar大鼠分为假手术组(S组)、缺血/再灌注组(Ⅰ组)和6%贺斯血液稀释组(H组),其中Ⅰ组和H组又各分为缺血10分钟再灌注2h、4h、8h和12h组,采用免疫组织化学方法检测脑组织ICAM-1的表达。结果Ⅰ组和H组再灌注2h、4h、8h和12h时ICAM-1的表达均较S组高(P均<0.01);H组再灌注2h、4h、8h和12h ICAM-1的表达均较Ⅰ组相应时间点低(P均<0.05)。结论(1)脑缺血/再灌注时血管内皮细胞ICAM-1的表达增加;(2)6%贺斯血液稀释降低再灌注早期阶段脑组织血管内皮细胞ICAM-1的表达。
Objective To investigate the effect of hemodilution of 6% hydroxyethyl starch (Hess, 200 / 0.5) on the expression of intercellular adhesion molecule-1 (ICAM-1) in rat brain after global cerebral ischemia / reperfusion. Methods Totally 84 Wistar rats were divided into three groups: sham operation group (S group), ischemia / reperfusion group (Ⅰ group) and 6% Hessian hemodilution group (H group) Blood was reperfused for 10 min at 2h, 4h, 8h and 12h, and the expression of ICAM-1 in brain tissue was detected by immunohistochemical method. Results The expression of ICAM-1 in group Ⅰ and group H at 2h, 4h, 8h and 12h after reperfusion was significantly higher than that in group S (all P <0.01). The expression of ICAM-1 at 2h, 4h, 8h and 12h after reperfusion in group H Compared with the corresponding time point in group I (P <0.05). Conclusions (1) The expression of ICAM-1 in vascular endothelial cells increased after cerebral ischemia / reperfusion; (2) 6% Hessian hemodilution reduced the expression of ICAM-1 in vascular endothelial cells in early stage of reperfusion.
