目的研究股神经肌支和隐神经纤维损伤7 d后二者蛋白表达的差异。方法成年雌性SD大鼠20只,体质量220~250 g。将大鼠右侧股神经干横断,左侧作为对照组不作处理。7 d后分别对正常股神经肌支及隐神经、损伤股神经肌支及隐神经取材,运用同位素标记相对和绝对定量技术标记(114标记正常的股神经肌支;115标记损伤的股神经肌支;116标记正常的隐神经;117标记损伤的隐神经)并结合二维液相色谱-串联质谱技术进行差异蛋白分析。结果共鉴定出3 358个蛋白。其中损伤的股神经肌支与正常的股神经肌支相比差异蛋白293个,表达升高112个,表达降低181个;损伤的隐神经神经与正常的隐神经相比差异蛋白417个,表达升高184个,表达降低233个。结论成功筛选出大鼠股神经运动神经纤维与感觉神经纤维损伤前后的差异蛋白,可为周围神经再生研究提供基础数据。
Objective To study the difference of protein expression between the nerve branch and the nerve fiber of the femoral nerve after 7 days. Methods Twenty adult female SD rats weighing 220-250 g were used. The right femur of rats were transected, and the left side was not treated as control group. After 7 days, normal nerve branch and saphenous nerve, injured femoral nerve branch and saphenous nerve were harvested respectively and labeled by relative and absolute quantification technique (114 normal femoral nerve branch; ; 116 marked normal saphenous nerve; 117 marked damaged saphenous nerve) and differential protein analysis combined with two-dimensional liquid chromatography-tandem mass spectrometry. A total of 3 358 proteins were identified. There were 293 differentially expressed proteins with 112 differentially expressed neurons in the injured femoral nerve branches, which were lower than the normal ones. There were 417 differentially expressed proteins in the lesioned saphenous nerve compared with the normal saphenous nerve Increased 184, decreased 233. Conclusion The successful screening of differentially expressed proteins of rat motor nerve fibers and sensory nerve fibers before and after injury can provide the basic data for the study of peripheral nerve regeneration.
