目的观察不同强度、不同作用时间的低频脉冲电磁场对C反应蛋白(CRP)作用下大鼠骨髓来源内皮祖细胞(EPC)增殖、凋亡及NO分泌能力的影响。方法密度梯度离心法获得SD雄性大鼠的骨髓EPC,实验分为10组,即空白对照组、12mg/LCRP组以及CRP+不同强度、不同作用时间(0.2mT2h、4h;0.6mT2h、4h;1.0mT2h、4h;1.4mT2h、4h)低频脉冲电磁场组。MTT法检测CPR对EPC增殖能力的影响,流式细胞仪检测CRP对EPC凋亡率的影响,硝酸还原酶法检测EPC培养液中NO含量的变化。结果与空白对照组相比,12mg/LCRP可明显抑制EPC增殖及NO分泌能力,促进EPC凋亡(P<0.05),0.6mT4h;1.0mT2h两组EPC增殖、NO分泌能力显著高于CRP组,EPC凋亡显著低于CRP组(P<0.05)。其余各组与CRP组相比无明显差异。结论12mg/LCRP可抑制EPC的增殖及NO分泌,促进其凋亡;0.6mT4h和1.0mT2h电磁场可拮抗CRP对EPC的作用,磁场对CRP环境下EPC的作用无强度及时间依赖性。
Objective To observe the effects of low frequency pulsed electromagnetic fields of different intensities and different time periods on the proliferation, apoptosis and NO secretion capacity of rat bone marrow-derived endothelial progenitor cells (EPCs) stimulated by C-reactive protein (CRP). Methods The bone marrow EPCs of SD male rats were obtained by density gradient centrifugation. The experiment was divided into 10 groups: blank control group, 12 mg / L CRP group and different intensity of CRP + for different duration of action (0.2mT2h, 4h; 0.6mT2h, 4h; 1.0mT2h , 4h; 1.4mT2h, 4h) low-frequency pulsed electromagnetic field group. The effect of CPR on EPC proliferation was detected by MTT assay. The effect of CRP on the apoptosis rate of EPC was detected by flow cytometry. The content of NO in EPC culture fluid was detected by nitrate reductase method. Results Compared with the blank control group, 12mg / L CRP significantly inhibited EPC proliferation and NO secretion, and promoted EPC apoptosis (0.6mT4h; 1.0mT2h EPC proliferation, NO secretion was significantly higher than the CRP group, EPC apoptosis was significantly lower than CRP group (P <0.05). There was no significant difference between the other groups and the CRP group. Conclusion 12 mg / L CRP can inhibit EPC proliferation and NO secretion and promote apoptosis. Electromagnetic fields of 0.6mT4h and 1.0mT2h can antagonize the effect of CRP on EPCs. The effects of magnetic field on EPC in CRP are not dependent on time and intensity.
