应用分子生物学技术研究白色念珠菌(以下简称白念)新基因型(C型和D型)的特征,与都柏林念珠菌(C.dubliniensis)和Ⅰ型星型念珠菌的标准菌株作比较,以美国全国临床实验室标准委员会(National Committeefor Clinical Laboratory Standards,NCCLS)M27-A法比较各菌株对氟康唑、5-氟胞嘧啶敏感性的差异。 材料和方法 供分析的念珠菌457株,包括白念439株,都柏林念珠菌5株,标准菌株13株。常规方法抽提细胞DNA,PCR反应体系为50μl终反应体积。设计三对引物,得到rDNA三个不同区域的PCR产物。其中两种扩增产物即基因间转录间隔序列(intergenic tran-scribed spacer,ITS)区和V3区rDNA分别经Dde Ⅰ、HaseⅢ
The characteristics of new genotypes (C and D) of Candida albicans (C. albicans) were studied by molecular biology techniques and compared with those of C. dubliniensis and C. albicans type I strains. The sensitivity of each strain to fluconazole and 5-fluorocytosine was compared with the National Committee for Clinical Laboratory Standards (NCCLS) M27-A method. Materials and Methods Candida 457 strains were analyzed, including 439 white yeast, 5 Candida albicans and 13 standard strains. Cell DNA was extracted by conventional methods. The final reaction volume was 50μl. Three pairs of primers were designed to obtain PCR products from three different rDNA regions.
